What is it?
A 5-laser spectral flow cytometer with 64 detection channels and 3 scatter parameters. Spectral flow cytometry enables ultra-high flow cytometry via spectral unmixing of spectral signatures to distinguish different fluorophore dyes and fluorescent proteins. Current literature has demonstrated the ability to detect up to 40 fluorescent parameters simultaneously. Also capable of nanoparticle detection (e.g. extracellular vesicles) with the Violet side scatter detector.

Instrument Overview: The Cytek Aurora is a 5-laser (Blue, Yellow Green, Red, Violet and UV) benchtop analyzer which uses APD detectors. It has three scatter channels, and 64 fluorescence detector channels, which allows the Cytek Aurora to collect the full spectrum of light emitted from stained samples and controls to create a spectral signature from the bound fluorophores. The acquisition software Spectroflo can unmix the spectral data in real time to distinguish similar fluorophores such as APC and Alexa-Fluor 647, thus increasing the number of cell markers that can be detected at the same time. This allows for simultaneous measurements of many surface and intracellular cell markers, enabling broad or deep phenotyping in a single sample. In addition, the detectors in the Cytek Aurora has a large dynamic range (7-decade display) which enables the detection of dim and bright markers in the same sample.
The 5-laser Cytek Aurora can detect up to 40 fluorescent parameters simultaneously, depending on the quality of the panel design (see OMIP-069). Panel design services are available in the SIgN Flow Cytometry platform.
With the addition of a Violet side scatter channel, the Cytek Aurora can also detect nanoparticles such as Extracellular Vesicles (EVs).
